ABSTRACT

Introdução: Espécies da família Asteraceae são conhecidas por apresentarem propriedades aromática, cosmética e terapêutica; tendo diversas pesquisas que evidenciou potencial medicinal dessa família. Dentre as espécies de Asteraceae, está Praxelis clematidea, que é rica em substâncias químicas como flavonoides, terpenóides e esteroides, as quais podem desempenhar uma série de atividades biológicas. Objetivo: Verificar o potencial tóxico do extrato etanólico das folhas de P. clematidea frente à células sanguíneas humanas, afim de determinar a toxicidade teórica dessa espécie. Métodos: Para a realização do teste de atividade citotóxica foram preparadas suspensões sanguíneas dos tipos A, B e O, que posteriormente foram misturadas a concentrações distintas do extrato etanólico por 1 (uma) hora. A hemólise foi quantificada por espectrofotometria em comprimento de onda de 540 nm. Resultados: O extrato etanólico das folhas de P. clematidea em diferentes concentrações apresentou baixa citoxicidade contra os eritrócitos humanos in vitro, enfatizando o produto como uma possível opção viável para a indústria de medicamentos fitoterápicos. No entanto, é importante elucidar que mais estudos in vivo precisam ser realizados para confirmar esse perfil toxicológico do extrato.


SUMMARY Introduction: Species of the Asteraceae family are known to have aromatic, cosmetic and therapeutic properties; having several researches that evidenced medicinal potential of this family. Among the species of Asteraceae, there is Praxelis clematidea, which is rich in chemical substances such as flavonoids, terpenoids and steroids, which can perform a series of biological activities. Aim: To verify the toxic potential of the ethanolic extract of P. clematidea leaves against human blood cells, in order to determine the theoretical toxicity of this species. Method: For the performance of the cytotoxic activity test, blood suspensions of types A, B and O were prepared, which were subsequently mixed at different concentrations of the ethanolic extract for 1 (one) hour. Hemolysis was quantified by spectrophotometry at a wavelength of 540 nm. Results: The ethanolic extract of P. clematidea leaves in different concentrations showed low cytotoxicity against human erythrocytes in vitro, emphasizing the product as a possible viable option for the herbal medicine industry. However, it is important to clarify that more in vivo studies need to be carried out to confirm this toxicological profile of the extract.


Introducción: Se sabe que las especies de la familia Asteraceae tienen propiedades aromáticas, cosméticas y terapéuticas; habiendo varias investigaciones que evidenciaron el potencial medicinal de esta familia. Entre las especies de Asteraceae, se encuentra Praxelis clematidea, que es rica en sustancias químicas como flavonoides, terpenoides y esteroides, que pueden realizar una serie de actividades biológicas. Objetivo: Verificar el potencial tóxico del extracto etanólico de hojas de P. clematidea contra las células sanguíneas humanas, con el fin de determinar la toxicidad teórica de esta especie. Metodos: Para realizar la prueba de actividad citotóxica se prepararon suspensiones de sangre de los tipos A, B y O, que luego se mezclaron a diferentes concentraciones del extracto etanólico durante 1 (una) hora. La hemólisis se cuantificó mediante espectrofotometría a una longitud de onda de 540 nm. Resultados: El extracto etanólico de hojas de P. clematidea a diferentes concentraciones mostró baja citotoxicidad contra eritrocitos humanos in vitro, destacando el producto como una posible opción viable para la industria de la fitoterapia. Sin embargo, es importante aclarar que es necesario realizar más estudios in vivo para confirmar este perfil toxicológico del extracto.

RESUMEN Introducción: Las enfermedades crónicas no transmisibles (ECNT) tienen las tasas de mortalidad y morbilidad más altas del mundo. El uso de plantas medicinales en la terapia de estas enfermedades es una realidad muy extendida. Sin embargo, la mayoría de las especies vegetales contienen cristales de oxalato cálcico, producto del metabolismo vegetal y su ingesta se correlaciona con la aparición de problemas renales como la litiasis urinaria, especialmente en personas predispuestas a estas afecciones. Objetivo: Determinar la cantidad de ácido oxálico en especies vegetales que se utilizan en el tratamiento de las ECNT. Metodología: Se obtuvieron extractos acuosos y ácidos de 0,5 g del fármaco vegetal de la especie y, posteriormente, se tituló con una solución estandarizada de permanganato de potasio 0,02 mol.L-1con las concentraciones de ácido oxálico expresadas en g/100g de fármaco vegetal seco. Resultados: La concentración de ácido oxálico osciló entre 4,58 ± 0,09 g/100 g a 17,21 ± 0,07 g/100 g en especies de plantas y la extracción de ácido fue más eficiente. Se realizó una optimización metodológica para las especies que presentaron los mejores resultados, Psidiumguajava y Artocarpus heterophyllus. Conclusión: Los datos obtenidos pueden servir de base para decisiones médicas y para profesionales que prescriben plantas medicinales.


SUMMARY Introduction: Chronic non-communicable diseases (CNCDs) are a group of diseases characterized by having the highest mortality and morbidity rates in the world. Despite the increase in new drug technologies, the use of medicinal plants as an aid in therapy for these diseases is a widespread reality. However, most plant species contain inorganic crystals of calcium oxalate, a product of plant metabolism, which has several functions in plant tissues. For the human species, its ingestion is associated with the arising of kidney problems such as urinary lithiasis, especially in people who have a predisposition to these conditions. Aim: To determine the amount of oxalic acid in plant species, which are used in the treatment of CNCDs. Methodology: After collection and characterization the plant species, aqueous and acidic extracts were obtained from 0.5 g of the plant drug of each species and, subsequently, were titrated with a standardized 0.02 mol.L-1 potassium permanganate solution and the concentrations of oxalic acid were expressed in g/100 g of dry vegetable drug. Result: The data obtained from the concentration of oxalic acid ranged from 4.58 ± 0.09 g/100 g to 17.21 ± 0.07 g/100 g and demonstrated that the concentrations from acid extraction are higher compared to the aqueous extraction, for all vegetables species analyzed. Methodological optimization was performed for the species that showed the highest results, Psidium guajava and Artocarpus hetero phyllus. Conclusion: The data obtained can serve as input for medical decisions and for professionals who prescribe medicinal plants.


RESUMO Introdução: As doenças crônicas não transmissíveis (DCNT) são um grupo de doenças caracterizadas por apresentar as maiores taxas de mortalidade e morbidade do mundo. Apesar do aumento de novas tecnologias medicamentosas, o uso de plantas medicinais como auxiliar na terapia dessas doenças é uma realidade bastante difundida. No entanto, a maioria das espécies vegetais contém cristais inorgânicos de oxalato de cálcio, um produto do metabolismo vegetal, que possui diversas funções nos tecidos vegetais. Para a espécie humana, sua ingestão está associada ao surgimento de problemas renais como a litíase urinária, principalmente em pessoas com predisposição a essas condições. Objetivo: Determinar a quantidade de ácido oxálico em espécies vegetais, que são utilizadas no tratamento de DCNT. Metodologia: Após a coleta e caracterização das espécies vegetais, extratos aquosos e ácidos foram obtidos a partir de 0,5 g da droga vegetal de cada espécie e, posteriormente, titulados com solução padronizada de permanganato de potássio 0,02 mol.L-1 e as concentrações de ácido oxálico foram expressos em g/100 g de fármaco vegetal seco. Resultado: Os dados obtidos da concentração de ácido oxálico variaram de 4,58 ± 0,09 g/100 g a 17,21 ± 0,07 g/100 g e demonstraram que as concentrações da extração ácida são maiores em relação à extração aquosa, para todas as espécies vegetais analisadas. A otimização metodológica foi realizada para as espécies que apresentaram os maiores resultados, Psidiumguajava e Artocarpus heterophyllus. Conclusão: Os dados obtidos podem servir de subsídio para decisões médicas e para profissionais que prescrevem plantas medicinais.

SUMMARY Introduction: biofilm-related infections caused by Staphylococcus aureus, Klebsiella pneumoniae and Pseudomonas aeruginosa are difficult to treat and few effective pharmacological options are currently available for this purpose. In this context, coumarin (2H-1-Benzopyran-2-one) has been reported to have antibacterial and antibiofilm activity, but this potential remains poorly understood. Aim: to investigate the action of coumarin on planktonic and biofilm forms of S. aureus, K. pneumoniae and P. aeruginosa. Results: a minimum inhibitory concentration (MIC) of coumarin ranging from 256 to 1024 fig/mL was observed, with a remarkable ability to inhibit the formation of biofilms and to act on mature biofilms in concentrations close to MIC. Conclusion: coumarin has strong activity against planktonic and biofilm forms on the three species of great relevance in the clinical scenario. These results are interesting to enable a pharmacological alternative for the treatment of these infections.


Introducción: las infecciones relacionadas con la biopelícula causadas por Staphylococcus aureus, Klebsiella pneumoniae y Pseudomonas aeruginosa son difíciles de tratar y actualmente existen pocas opciones farmacológicas eficaces para este propósito. En este contexto, se ha informado que la cumarina (2H-1-Benzopiran-2-ona) tiene actividad antibacteriana y antibiofilm, pero este potencial sigue siendo poco conocido. Objetivo: investigar la acción de la cumarina sobre formas planctónicas y de biopelículas de S. aureus, K. pneumoniae y P. aeruginosa. Resultados: se observó una concentración inhibitoria mínima (CMI) de cumarina en el rango de 256 a 1024 µg/mL, con una notable capacidad para inhibir la formación de biofilms y actuar sobre biofilms maduros en concentraciones cercanas a la CMI. Conclusión: la cumarina tiene una fuerte actividad contra las formas planctónicas y biofilm sobre las tres especies de gran relevancia en el escenario clínico. Estos resultados son interesantes para habilitar una alternativa farmacológica para el tratamiento de estas infecciones.

Objetivo: investigar a ação da cumarina sobre as formas planctónicas e de biofilme de S. aureus, K. pneumoniae e P. aeruginosa. Resultados: observou-se uma concentração inibitória mínima (CIM) de cumarina variando de 256 a 1024 µg/mL, com notável capacidade de inibir a formação de biofilmes e de atuar sobre biofilmes maduros em concentrações próximas à CIM. Conclusão: a cumarina possui forte atividade contra as formas planctónicas e de biofilme sobre as três espécies de grande relevância no cenário clínico. Esses resultados são interessantes para possibilitar uma alternativa farmacológica para o tratamento dessas infecções.

ABSTRACT

Diabetes Mellitus (DM) is a metabolic syndrome characterized by hyperglycemia. Chronic complications affect a number of organs, including the lungs. Cissampelos sympodialis Eichl (Menispermaceae) is a plant used to treat respiratory diseases. The aim of this study was to evaluate the effect of Cissampelos sympodialis extract (CSE) in lungs of diabetic rats. We used 30 Wistar rats divided into three groups: control group (CG), diabetic group (DG) and diabetic Cissampelos sympodialis treatment group (DTG). Diabetes was induced by streptozotocin (40 mg/kg i.v.). The CSE (400 mg/kg, po) was administered daily, during four weeks, beginning one week after the onset of DM. The treatment with CSE was not able to reduce blood glucose levels after streptozotocin injection. However, it was able to decrease cholesterol and triglycerides and prevent damage on pancreatic islets morphology. Additionally, morphological alterations such as alveolar septa loss, inflammatory infiltrate and fibrosis were seen in lung tissue of rats with DM, and treatment with CSE apparently reversed these histopathological findings. Thus, CSE treatment reduced the lipid profile and restored the lung architecture of diabetic animals by a mechanism independent of glycemia and which might be associated with the reduction of the damage on the pancreatic islets.

Abstract Fungal infections have become a concern for health professionals, and the emergence of resistant strains has been reported for all known classes of antifungal drugs. Among the fungi causing disease, we highlight those that belong to the genus Aspergillus. For these reasons, the search for new antifungals is important. This study examines the effects of a coumarin derivative, 4-acetatecoumarin (Cou-UMB16) both alone and together with antifungal drugs, and its mode of action against Aspergillus spp. Cou-UMB16 was tested to evaluate its effects on mycelia growth, and germination of Aspergillus spp. fungal conidia. We investigated its possible action on cell walls, on the cell membrane, and also the capacity of this coumarin derivative to enhance the activity of antifungal drugs. Our results suggest that Cou-UMB16 inhibits Aspergillus spp. virulence factors (mycelia growth and germination of conidia) and affects the structure of the fungal cell wall. When applying Cou-UMB16 in combination with azoles, both synergistic and additive effects were observed. This study concludes that Cou-UMB16 inhibits mycelial growth and spore germination, and that the activity is due to its action on the fungal cell wall, and that Cou-UMB16 could act as an antifungal modifier.

ABSTRACT Cissampelos sympodialis Eichler, Menispermaceae, a Brazilian medicinal plant and its alkaloid warifteine present immunomodulatory activity on asthma experimental model by reducing antigen-specific IgE levels, eosinophil infiltration and lung hyperactivity. Allergic rhinitis is a chronic inflammatory disorder of the nasal tissue that affect the quality of life and it is a risk factor for asthma exacerbation. This study evaluated the effect of inhaled warifteine in an allergic ovalbumin rhinitis model. Inhaled warifteine (2 mg/ml) treatment of ovalbumin-sensitized BALB/c mice significant decreased total and differential number of cells on the nasal cavity and decreased ovalbumin-specific IgE serum levels. Hematoxylin & eosin staining of histological preparations of ovalbumin nasal tissues showed changes such as congestion and a massive cell infiltration in the perivascular and subepithelial regions characterizing the nasal inflammatory process. However, inhaled warifteine or dexamethasone treatment decreased cell infiltration into the perivascular regions and it was observed an intact nasal tissue. Periodic acidic staining of nasal epithelium of ovalbumin animals demonstrated high amount of mucus production by goblet cells and inhaled warifteine or dexamethasone treatment modulated the mucus production. In addition, toluidine blue staining of the nasal epithelium of ovalbumin animals demonstrated an increase of mast cells on the tissue and inhaled warifteine or dexamethasone treatment decreased in average of 1.4 times the number of these cells on the nasal epithelium. Taken these data together we postulate that warifteine, an immunomodulatory alkaloid, can be a medicinal molecule prototype to ameliorate the allergic rhinitis conditions.

Abstract In a previous study, we reported the antispasmodic and gastroprotective effects of the Serjania caracasana (Jacq.) Willd., Sapindaceae, extract. In the present study, we evaluated the LD50, hemolytic and antispasmodic activities of its fractions and characterized its major constituents by isolation and GC-MS. The animals showed non-toxic symptoms with oral doses up to 2000 mg/kg, suggesting a safe oral administration. Furthermore, a low hemolytic activity was detected for the saponin fraction. Antispasmodic activity of the fractions was evaluated through carbachol-induced contractions in rat ileum. The hexane fraction was the most potent (IC50 68.4 ± 5.9 µg/ml) followed by the dichloromethane fraction (IC50 161.3.4 ± 40.7 µg/ml). Butanol fraction was the less effective (IC50 219.8 ± 60.3 µg/ml). The phytochemical study of the S. caracasana fractions afforded the isolation of friedelin, β-amyrin, allantoin and quercitrin. This is the first time that the presence of allantoin and quercitrin in the Serjania genus has been reported. Among the isolated compounds and those characterized by GC-MS, β-amyrin and β-sitosterol were present in the most active fractions, hexane and dichloromethane, and they may be related to its antispasmodic activity. In addition, spathulenol was only found in the hexane fraction and its presence might justify the highest antispasmodic activity observed for this fraction.

ABSTRACT INTRODUCTION: Schistosomiasis, a parasitic disease caused by trematode flatworms of the genus Schistosoma, affects more than 200 million people worldwide, and its control is dependent on a single drug, praziquantel. Here, we report the in vitro effect of rotundifolone, a monoterpene isolated from Mentha x villosa (Lamiaceae), on Schistosoma mansoni adult worms. METHODS: The in vitro effect of rotundifolone on adult Schistosoma mansoni was evaluated by analysis of behavior and mortality and through a scanning electron microscopic analysis of ultrastructural changes in the tegument of the worms. RESULTS: At concentrations of 3.54 and 7.09μg/mL-1 rotundifolone, no worm mortality was observed at any of the sampling intervals. A minor reduction in movement of the tail, suckers, and gynecophoral canal membrane was observed after 96 h of exposure to 7.09μg/mL-1 rotundifolone. At 70.96μg/mL-1, a lack of movement was observed from 72h onwards and all worms were deemed dead; similar effects were observed at 48h with 177.4μg/mL-1, and at 24h with 354.8μg/mL-1 and 700.96μg/mL-1. Rotundifolone also caused death of all parasites and separation of coupled pairs into individual males and females after 24h at 354.8μg/mL-1. CONCLUSIONS: The main changes in the tegument induced by the different ROT treatments were: after 24h incubation, bubble lesions spread over the entire body and loss of tubercles occurred in some regions of the ventral region.

ABSTRACT Dengue is the most important viral infection transmitted among humans by arthropod-borne. There are currently no vaccines or specific therapeutical treatment. Therefore, immunomodulatory compounds from plants have been widely examined for their antiviral effects. Cissampelos sympodialis Eichler, Menispermaceae, has scientifically proven to present immunomodulatory activities. Here we assessed the antiviral activity of leaf hydroalcoholic extract, warifteine or methylwarifteine from C. sympodialis in an in vitro dengue virus infection model. The results demonstrated that leaf hydroalcoholic extract or warifteine/methylwarifteine treatment did not reduce dengue virus-Ag+ hepatocyte (Huh-7 cell) rates in present experimental conditions. However, we assessed the potential antiviral effect of leaf hydroalcoholic extract or warifteine/methylwarifteine on dengue virus-infection by the production of inflammatory molecules, TNF-α, MIF, IL-8 and PGE2. Dengue virus infection enhanced TNF-α, MIF, IL-8 and PGE2 production in infected Huh-7 cells and leaf hydroalcoholic extract but not warifteine/methylwarifteine treatments, significantly reduced these molecules in infected cells. In dengue virus-infected Huh-7 cells, non-structural protein-1 is produced and leaf hydroalcoholic extract significantly inhibited it independently of alkaloids. Our findings imply that leaf hydroalcoholic extract may attenuate dengue virus infection in Huh-7 cells by inhibiting the enhanced of pro-inflammatory mediators and non-structural protein-1 production induce by dengue virus independently of warifteine/methywarifteine its major compound.

Introduction: The essential oil Mentha x villosa (MVEO) has a wide range of actions, including antibacterial, antifungal, antiprotozoal and schistosomicidal actions. The present study aimed to investigate the ultrastructural changes of MVEO on the tegument of adult Schistosoma mansoni. Materials and Methods: Different concentrations of MVEO were tested on S. mansoni adult worms in vitro. Ultrastructural changes on the tegument of these adult worms were evaluated using scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Results: The MVEO caused the death of all worms at 500 μg mL-1 after 24 h. After 24h of 500 μg mL-1 MVEO treatment, bubble lesions were observed over the entire body of worms and they presented loss of tubercles in some regions of the ventral portion. In the evaluation by TEM, S. mansoni adult worms treated with MVEO, 500 μg mL-1, presented changes in the tegument and vacuoles in the syncytial matrix region. Glycogen granules close to the muscle fibers were visible. Conclusion: The ability of MVEO to cause extensive ultrastructural damage to S. mansoni adult worms correlates with its schistosomicidal effects and confirms earlier findings with S. mansoni.

ABSTRACT

Abstract Natural marine products can help increase the quality of life in patients with neurological diseases. A large number of marine products act against Alzheimer's disease through varying pathways. According to structure- and ligand-based analyses, caulerpin, an alkaloid primarily isolated from the genus Caulerpa, possesses activity against monoamine oxidase B. To predict the activity of caulerpin, we employed Volsurf descriptors and the machine learning Random Forest algorithm in parallel with a structure-based methodology that included molecular docking. Using caulerpin as a lead compound, a database containing 108 analogs was evaluated, and nine were selected as active. The structures selected as active exhibited polar and non-polar substitutions on the caulerpin skeleton, which were relevant for their activity. Dragon consensus drug-like scoring was applied to identify the active analogs that might serve as good drug candidates, and the entire group presented satisfactory performance. These results indicate the possibility of using these analogs as potential leads against Alzheimer's disease.

Abstract Marine algae have been the focus of important studies over the past fifty years, with a considerable number of components important to chemists and taxonomists having been isolated and characterized. The scientific data available on Sargassum polyceratium are extremely limited. The objective of the present study was to evaluate the antinociceptive activity of an ethanol extract of S. polyceratium and to isolate its components. Intraperitoneal treatment with ethanol extract of S. polyceratium reduced the number of acetic acid-induced writhes and the amount of time spent in paw-licking in the second phase of the formalin test. Ethanol extract of S. polyceratium also reduced the amount of time spent in paw-licking in the glutamate test; however, there was no difference in the reaction time in the hot plate test at any of the doses tested. The chemical components isolated from ethanol extract of S. polyceratium were identified using one- and two-dimensional spectroscopic methods such as infrared spectroscopy, mass spectrometry and 1H and 13C nuclear magnetic resonance spectroscopy. The analytical results were also compared with data obtained in the literature. The following porphyrin derivatives were isolated from S. polyceratium: 132-hydroxy-(132-R)-pheophytin-a, 132-hydroxy-(132-S)-pheophytin-a, pheophytin-a, and the steroid fucosterol. The present results indicate that the ethanol extract of S. polyceratium has antinociceptive activity. In addition, four new substances were isolated from the species evaluated.

ABSTRACT

Este trabalho teve como objetivo avaliar o efeito de diferentes tipos de telas de sombreamento utilizadas como subcobertura no cultivo hidropônico de alface. Para isso, foi conduzido um experimento em estufas cobertas com filme plástico de 150µm com três tipos de subcobertura: tela termorrefletora, preta, vermelha e um ambiente controle sem o uso de tela. Para avaliar a modificação ambiental ocasionada pelos tratamentos, foram instalados em cada estufa e no ambiente externo um termohigrômetro data logger, que registrou a temperatura a cada 15 minutos do dia 18/05/2011 ao dia 14/06/2011. Posteriormente, foram calculados os graus-dia acumulados em cada ambiente. Para analisar as variações de crescimento das plantas nas diferentes condições de cultivo, foram realizadas cinco coletas ao longo do período experimental. Após a coleta das plantas, elas eram levadas ao laboratório para contagem do número de folhas maiores que 10cm de comprimento e para a determinação do índice de área foliar. Os resultados indicaram que o uso das telas alterou os padrões da temperatura, quando o uso das telas termorrefletora e preta reduziu a temperatura máxima em 4,7 e 3,6ºC, respectivamente, em relação ao ambiente controle, permitindo um menor acúmulo térmico no período experimental. Quanto às alterações morfológicas ocasionadas pelas modificações ambientais, foi possível constatar que o uso das telas termorrefletora e preta aumentou o número de folhas maiores que 10cm e o índice de área foliar. O uso de tela preta ou termorrefletora como subcobertura em estufas pode ser uma alternativa viável para incrementar a produção de alface em condições tropicais.


This study aimed to evaluate the effect of different types of shading nets used as undercover of plastic in greenhouses of hydroponic cultivation of lettuce. The experiment was performed in greenhouses covered with plastic film of 150µm with three types of undercover nets: thermal reflector net, black, red and acontroltreatment - without the use of the undercover.To evaluate the environmental changes caused by treatments thermohygrometer data logger swere installed in each greenhouse as well asone in the external environment. The devices recorded the air temperature every 15 minutes from May 18th2011to June 14th2011. By using these data, it was possible to estimate the values of the growing degree-daysineach environment. To analyze the changes in plant growth and development caused by the treatments, five samples were taken along the experimental period. Inthe laboratorywe counted the number of leaves greater than 10cmof lengthand measured the leaf area index. The results indicated that the use of the nets changed the overall patterns of temperature and relative humidity. The use of thermal reflector net and black net reduced the maximum temperature at 4.7 and 3.6°C, respectively, compared to the controlalso allowing for lower growing degree-days at the end of the experimental period.As for the morphological changes caused by treatments, we observed that the use of thermal reflector netand black net increased the number of leaves greater than 10cm andalso the leaf area index.The use of black net or thermal reflector net as undercover in greenhouses can be a feasible alternative to increase the production of lettuce in tropical conditions.

ABSTRACT

The flavonoids are a large class of polyphenolic compounds found in plants that are known to exhibit biological effects. In the study, the flavonoid 5.7.4’-trimethoxyflavone (TMF) extracted from Praxelis clematidea was evaluated for its antibacterial activity. Microdilution method was used for antibacterial assay of the flavonoid and eleven bacteria strains were used in the study for activities. The results were also compared with the standard drug, Chloramphenicol (100 ug/mL). The results obtained showed activity of the flavonoid against Gram positive and Gram negative bacteria.


Los flavonoides son una clase importante de compuestos polifenólicos encontrados en las plantas que se sabe que presentan efectos biológicos. En el estudio, el flavonoide 5.7.4 '-trimethoxyflavone (TMF) extraído de Praxelis clematidea fue evaluado por su actividad antibacteriana. Se utilizó el método de microdilución para el ensayo antibacteriano del flavonoide y once cepas de bacterias se usaron en el estudio de las actividades. Los resultados se compararon también con el fármaco estándar, Cloranfenicol (100 ug/mL). Los resultados obtenidos mostraron actividad del flavonoide contra bacterias Gram positivas y Gram negativas.

ABSTRACT

The cardiovascular effects elicited by the ethanolic extract obtained from the roots of Erythroxylum pungens O.E. Schulz, Erythroxylaceae (EEEP) and the vasorelaxant effect induced by its main tropane alkaloid (pungencine) were investigated. In normotensive rats, administration of EEEP (1, 10, 30 and 60 mg/kg i.v., randomly) produced dose-dependent hypotension (-2±1, -7±0.5 -17.6±1, -24±1 Δ mmHg, n=5) followed by tachycardia (3±0.5, 7±2, 7.1±1, 10±5 Δ bpm, n=5). In intact phenylephrine (Phe, 10 µM)-pre-contracted rings, EEEP (0.01-500 µg/mL) induced concentration-dependent vasorelaxation (EC50 13.7±5.5 µg/mL, Maximal Response= 92±2.6%), and this effect was unchanged after the removal of the vascular endothelium (EC50 27.2±4.7 µg/ml, Maximal Response= 88.3±3.3 %). In KCl (80 mM)-pre-contracted-endothelium-denuded rings, EEEP elicited concentration-dependent relaxation (EC50= 128.2±11.2 µg/mL, Maximal Response 76.8±3.4%). Vasorelaxation has also been achieved with tonic contractions evoked by the L-type Ca2+ channel agonist Bay K 8644 (EC50 80.2±9.1 µg/mL, Maximal Response 86.3±8.3%). In addition, in a depolarizing medium, EEEP inhibited CaCl2 (30-500 µg/mL) induced contractions and caused a concentration-dependent rightward shift of the relaxation curves. Lastly, the tropane alkaloid pungencine caused vasorelaxation in mesenteric arteries resembling to the EEEP responses. These results suggests that EEEP induces hypotension and vasorelaxation, at least in part, due to the reduction in [Ca2+]i in vascular smooth muscle cells.

INTRODUCTION: Resistant arterial hypertension may lead to muscle disuse and reduced functional capacity due to arterial and target-organs lesions. The main objective of this work is to evaluate the quadriceps strength and fatigue tolerance after a program of resistance exercise in subjects with resistant primary hypertension. METHODS: Six patients under pharmacological treatment were submitted to a four-week resistance exercise training program for the quadriceps (8-14 repetitions, 3 sets, 3 days per week). Strength was evaluated by isometric dynamometry, as the percentage change in maximum voluntary contraction over the four week program. Fatigue was analyzed by surface electromyography, as the change in both root mean square value and intercept of median frequency slope of vastus medialis and vastus lateralis. RESULTS: Significant increase in the maximum voluntary contraction was observed (p = 0.04). Fatigue tolerance was not improved as seen by root mean square as well as in the intercept of median frequency (p > 0.05). Additionally, no significant changes were observed in resting arterial blood pressure and heart rate throughout the training period. CONCLUSION: The prescribed protocol seemed to successfully increase localized muscle strength without negatively affecting the monitored cardiovascular variables in patients with resistant hypertension under pharmacological treatment.


INTRODUÇÃO: A hipertensão arterial resistente pode levar ao desuso muscular e redução da capacidade funcional devido a lesões arteriais e de órgãos-alvo. O objetivo deste trabalho é avaliar a força muscular e resistência à fadiga do quadríceps em indivíduos com hipertensão primária resistente após um programa de exercícios de resistência. MÉTODOS: Seis pacientes sob tratamento farmacológico foram submetidos a um programa de treinamento muscular do quadríceps por 4 semanas (8-14 repetições, 3 séries, 3 dias por semana). Força foi avaliada por meio de dinamometria isométrica e apresentada como o percentual de alteração na contração voluntária máxima ao longo do programa de treinamento. Fadiga foi analisada por meio da eletromiografia de superfície como a alteração tanto na raiz quadrática média e no ponto de interceptação da inclinação da frequência mediana dos músculos vasto lateral e medial. RESULTADOS: Foi observado aumento significante na força isométrica máxima (p = 0,04). A tolerância à fadiga não foi alterada, como evidenciado pela raiz quadrática média ou interceptação da frequência mediana (p > 0,05). Adicionalmente, não foram observadas alterações significativas na pressão arterial de repouso e frequência cardíaca ao longo do programa. CONCLUSÃO: O protocolo prescrito aumentou a força muscular localizada sem alterar negativamente as variáveis cardiovasculares monitoradas nos pacientes com hipertensão resistente sob tratamento farmacológico.

ABSTRACT

This work studied the anti-inflammatory activities of the hydroalcoholic extracts (HAEs) from Erythrina velutina Willd. (Ev) and E. mulungu Mart. ex Benth. (Em) in the carrageenan- and dextran-induced mice hind paw edema models. These medicinal plants belonging to the Fabaceae family are used in some Brazilian communities to treat pain, inflammation, insomnia and disorders of the central nervous system. In the present work, the extracts were administered orally in male mice at the doses of 200 or 400 mg/kg. In the carrageenan-induced test, only Em showed anti-inflammatory activity, decreasing the paw edema, at the doses of 200 and 400 mg/kg. No effect was observed with Ev in this model. On the other hand, in the dextran model, Ev demonstrated anti-inflammatory effect, showing decrease of the paw edema at the 1, 2, 3, 4 and 24th h. Em (200 or 400 mg/kg) presented anti-inflammatory effect at the 2, 3 and 4th h after administration of dextran, as compared to control. In conclusion, the work showed that Ev and Em present anti-edematous actions, which possibly occurs by distinct mechanisms. While Ev seems to interfere especially in inflammatory processes in which mast cells have an important role, Em exerts greater activity in the inflammatory process that depends mainly on polymorphonuclear leucocytes. However, further studies are needed to determine the exact mechanism of action of the species investigated.

ABSTRACT

An atisane diterpene was isolated from Xylopia langsdorfiana St. Hilaire & Tulasne, Annonaceae, leaves, ent-atisane-7α,16α-diol (xylodiol). Preliminary study showed that xylodiol was cytotoxic and induced differentiation on human leukemia cell lines. However, the molecular mechanisms of xylodiol-mediated cytotoxicity have not been fully defined. Thus, we investigated the anti-tumor effect of xylodiol in human leukemia HL60 cell line. Xylodiol induced apoptosis and necrosis. HL60 cells treated with xylodiol showed biochemical changes characteristic of apoptosis, including caspases-8, -9 and -3 activation and loss of mitochondrial transmembrane potential (∆ Ψm). However, there was a condensation rather than swelling of mitochondria. Moreover, the formation of condensed mitochondria and the loss of ∆ Ψm occurred downstream of caspase activation. Cyclosporine A did not protect HL60 cells from the cytotoxic effects of xylodiol, suggesting that the loss of ∆ Ψm is a late event in xylodiol-induced apoptosis. Oxidative stress was involved in xylodiol-induced apoptosis. Thus, we conclude that activated caspases cleave cellular proteins resulting in mitochondrial damage leading to mitochondrial condensation, loss of ∆ Ψm and ROS release from the mitochondria. ROS can further induce and maintain a collapse of ∆ Ψm leading to cellular damage through oxidation of lipids and proteins resulting in apoptotic cell death.

ABSTRACT

Concerning the reduction of preslaughter death losses, high temperature and relative humidity in the tropics are the major concerns regarding broiler survival prior slaughtering. However, the relationship between different lairage times under controlled environment and the thermal condition outside the holding area is still unclear. Thus, the aim of this study was to compare different intervals of lairage time with different environmental temperature and relationship with poultry mortality rates. A study was conducted in a commercial poultry abattoir in the State of Sao Paulo, Brazil, in the year of 2006. The historical data set from 13,937 trucks, about broiler mortality during preslaughter operations, was given by the abattoir. Factors that influence the welfare of birds were studied, such as lairage time in abattoir and hourly dry-bulb temperature. The statistical analysis was performed using the Double Generalized Linear Models. The lower incidence of death losses before arrival at the processing plant (approximately 13 dead birds per truck) was observed when the lairage time was between 1 to 3 hours, under high temperatures (above 22°C). This effect was more pronounced under critical (25-28°C) and lethal intervals (above 29°C) (12 and 13 dead birds per truck, respectively). In relation to lairage time, the reduction in mortality rates was more pronounced in the intervals up to 1 hour of lairage under climatized conditions, with a reduction of 3 dead birds per truck. It was concluded that thermal stress have negative influence for broilers welfare kept under preslaughter conditions and variations on mortality rate are strongly related with different lairage times.


No que diz respeito à redução das perdas por mortalidade pré-abate, as temperaturas e umidades relativas elevadas nos trópicos são as maiores preocupações quanto à sobrevivência de frangos de corte antes do abate. Entretanto, a relação entre diferentes tempos de espera em ambiente controlado e a condição térmica fora do galpão de espera ainda não está clara. Dessa forma, objetivou-se por meio deste trabalho comparar diferentes tempos de espera com diferentes temperaturas externas e a influência nas taxas de mortalidade de frangos de corte. O estudo foi conduzido em um abatedouro comercial de frangos de corte, situado no Estado de São Paulo, Brasil, durante o ano de 2006. Dados históricos de mortalidade de aves durante as operações pré-abate foram obtidos no abatedouro, provenientes de 13.937 caminhões transportadores de frangos de corte. Fatores que influenciam o bem estar das aves foram estudados, dentre eles, o tempo de espera no abatedouro e a temperatura horária do ambiente externo. A análise estatística foi realizada por meio dos Modelos Lineares Generalizados Duplos. A baixa incidência de mortes antes da chegada à linha de abate foi observada (aproximadamente 13 aves mortas por caminhão) quando o tempo de espera esteve entre 1 e 3 horas, sob altas temperaturas (acima de 22°C). Esse efeito foi mais evidente nas faixas crítica (25-28°C) e letal (acima de 29°C) (12 e 13 aves mortas por caminhão, respectivamente). Com relação ao tempo de espera, a redução da mortalidade foi mais pronunciada em intervalos acima de uma hora de espera, sob condições de espera climatizada. Conclui-se que o estresse térmico possui influência negativa para o bem-estar de frangos mantidos sob condição pré-abate e variações na taxa de mortalidade são fortemente relacionadas aos diferentes tempos de espera.

ABSTRACT

The wood as a source of organic material responds in different ways to different treatments her employees, including the application of glue which depends on its moisture content, tending to reach a moisture content in dynamic equilibrium with relative humidity of the atmosphere. Currently we use blades with moisture levels of between 5 and 15% according to the type of resin used. This study evaluated the effect of 04 weights (270, 330, 364 and 390 g/m²) in the production of panels using offset the species: Copaifera ducker Dawyer and Eperua oleifera Ducke. The species were removed from the area of sustainable forest management Gethal Plywood Industry Ltd. located in Manicoré-Am. The slides were produced with thickness of 2.2 mm. The variables of the pressing cycle of were controlled by following the technical guidelines set by the manufacturer of resin. The property was assessed to resilience of the line of glue, with samples of the hood and the kernels in drought conditions and post-boil. Statistical analysis was applied an experimental design with in a factorial arrangement of treatments. Confronting the average values of voltage collapse and the percentage of failure in wood obtained in this research, test and post-boiling dry, with the literature, it appears that the panels assessed meet the established criteria, may be indicated for use inside and outside Because according to EN 314-2 (1993) the average voltage collapse along with the percentage of failure in the wood are at an acceptable standard of behavior.


A madeira por ser um material de origem orgânica responde de várias maneiras aos diversos produtos nela empregados, inclusive a aplicação de cola a qual depende do seu teor de umidade, tendendo a atingir um teor de umidade em equilíbrio dinâmico com a umidade relativa da atmosfera. Atualmente utilizam-se lâminas com teores de umidade entre 5 e 15% de acordo com o tipo de resina utilizada. O presente estudo avaliou o efeito de 04 gramaturas (270; 330; 364 e 390 g/m²) na produção de painéis compensados utilizando as espécies: Copaifera duckei Dawyer e Eperua oleifera Ducke. As espécies utilizadas foram retiradas da área de manejo florestal sustentável da Gethal Indústria de Madeira Compensada Ltda localizada em Manicoré-Am. As lâminas foram produzidas com espessuras de 2,2 mm. As variáveis do ciclo de prensagem foram controladas seguindo as orientações técnicas estabelecidas pelo fabricante da resina. Foi avaliada a resistência da linha de cola, com amostras da capa e do miolo em condições seca e pós-fervura. Para análise estatística foi aplicado um delineamento inteiramente casualizado com arranjo em esquema fatorial dos tratamentos. Confrontando os valores médios da tensão de ruptura e a percentagem de falha na madeira obtidos nesta pesquisa, ensaio seco e pós-fervura, com os da literatura, foi verificado que os painéis avaliados atendem aos critérios estabelecidos, podendo ser indicados para uso interior e exterior, pois de acordo com a Norma EN 314-2 (1993) o valor médio da tensão de ruptura em conjunto com a porcentagem de falha na madeira encontram-se em um padrão aceitável de comportamento.
